Characterization of sHRP and CLuc. Protein sequence schematics and molecular weight analysis. A HRP was driven by CMV promoter with a myc-tag and a SP for secretion. Supernatants and whole cell lysate from transfected cells were collected and analyzed with Western blotting. B CLuc was engineered with a similar sequence design to HRP. 293 T-sHRP; HEK- 293 T cells transiently transfected with sHRP, 293 T (NC); untransfected 293 T cells as a negative control (NC). Actin was used as a loading control. C-E serum-containing media groups, F–H SFM groups; untransfected 293 T cells served as a negative control. ( and ) sHRP-ABTS, ( and ) sHRP-TMB, ( and ) CLuc-vargulin. All error bars show SD; all x-axes were converted into exponential of 2, n = 3, with 3 independent repeats
